Abstract

Analisis viabilitas bakteri Escherichia coli penting untuk memahami respons sel terhadap stres termal, khususnya terkait mekanisme kerusakan membran dan kematian sel. Penelitian ini bertujuan menganalisis karakteristik kematian bakteri Escherichia coli akibat paparan stres termal menggunakan pendekatan multimodal yang mengintegrasikan parameter elektrik dari Spektroskopi Impedansi Listrik (SIL) dan optik dari spektrofotometri OD260 melalui model Multiple Linear Regression (MLR). Penelitian menggunakan satu kultur bakteri yang dibagi menjadi empat kelompok perlakuan suhu (37°C, 45°C, 55°C, dan 65°C) selama 5 menit. Pengukuran SIL dan OD260 dilakukan sebanyak tiga kali pada setiap kelompok perlakuan, sedangkan metode Total Plate Count (TPC) digunakan sebagai ground truth. Data dianalisis menggunakan Google Colaboratory (Google Colab) melalui regresi linear sederhana untuk menentukan frekuensi impedansi terbaik, kemudian dikembangkan menjadi model regresi linear berganda dan divalidasi menggunakan nilai koefisien determinasi (R²) dan Root Mean Square Error (RMSE). Hasil penelitian menunjukkan bahwa peningkatan suhu menurunkan populasi bakteri hidup yang diikuti oleh peningkatan nilai OD260 dan penurunan nilai impedansi. Model parameter tunggal menghasilkan nilai R² sebesar 0,9784 untuk OD260 dan 0,9180 untuk impedansi, sedangkan model multimodal meningkatkan performa menjadi R² = 0,9936 dengan RMSE = 0,1329. Peningkatan nilai R² dan rendahnya RMSE menunjukkan bahwa integrasi parameter elektrik dan optik melalui regresi multimodal meningkatkan akurasi prediksi dibandingkan model parameter tunggal dalam mengidentifikasi fase kematian bakteri secara presisi.
